Association of the transmembrane TGF-a precursor with a protein kinase complex

, : Association of the transmembrane TGF-a precursor with a protein kinase complex. The Journal of Cell Biology 125(4): 3-16

Two cellular proteins that interact with transmembrane transforming growth factor-a (TGF-a) were identified. Like a variety of other growth factors, TGF-a is synthesized as a transmembrane precursor; the protein's short, cysteine-rich cytoplasmic domain does not contain any known functional motifs. An 86 kDa protein (p86) and a tyrosine-phosphorylated 106 kDa protein (p106) were found to form a complex with protein kinase activity that required the cytoplasmic domain of transmembrane TGF-a. The presence of this interaction suggests that the protein kinase activity of the complex may be regulated by ligand-receptor interaction. Therefore, in relation to direct cell-cell contact, the interaction between transmembrane TGF-a and its receptor may form the basis of a 2-directional communication system. .
