The expression and purification of rhodopsin kinase (RK) are reported. The expression of the RK gene in the yeast Pichia pastoris, COS 1 cells, and an HEK293 stable cell line was compared. The use of the pCMV5 vector to express RK in COS cells was the most satisfactory method, and the expressed enzyme with an N-terminal histidine tag was purified using a 2-step protocol. The purified enzyme demonstrated correct posttranslational modifications and a somewhat broader pH vs. catalytic activity profile in comparison with the wild-type enzyme.
