Identification and virulence determination of cytotoxigenic E. coli strains by conventional methods and PCR

, : Identification and virulence determination of cytotoxigenic E. coli strains by conventional methods and PCR. Medycyna Weterynaryjna 55(2): 129-133

The pathogenic strains of Verocytotoxin-producing E. coli (VTEC) are a real epidemiological threat for humans and animals. It is very difficult to identify these virulent strains, so it very important to work out diagnostic methods which will detect these pathogens in the environment quickly and effectively. In the present paper the virulence properties of 8 E. coli strains were analyzed. Determination of heat-labile enterotoxin 1, and verocytotoxins VT1 (SLT-II) and VT2 (SLT-II) was accomplished in the reversed passive latex agglutination (RPLA). E. coli strains were also tested for verocytotoxin production using an amplification of SLT-IA and SLT-IIA genes by the PCR procedure. Its ability to decompose urea and sorbitol (in order to identification of 01-57:H7 E. coli strains) was defined as well as its capacity to produce haemolysin. Biochemical tests of PCR reaction and RPLA kits proved also useful to identify pathogens belonging to Verocytotoxin-producing and enterotoxigenic Escherichia coli strains. These tests save time and give appropriate diagnostic efficiency.
