AIM: To study the electrochemical behavior and reaction mechanism of rufloxacin hydrochloride (RUFX) in phosphate buffer solution. A method of Adsorptive Stripping Voltammetry for the determination of RUFX was established. METHODS: Linear-Sweep polarography, cyclic Voltammetry and direct current polarography were used in the present research. RESULTS: In a supporting electrolyte containing KH2PO4-Na2HPO4 (pH7.00), a reduction peak of RUFX was observed by linear-sweep voltammetry at Hg electrode. The peak shows a potential of -1.27V (vs. Ag/AgCl) and adsorptive characteristics. The adsorbed species is most probably the neutral molecular of RUFX. The saturated adsorption amount of RUFX at Hg electrode is 5.51X10-11mol/cm2 and every RUFX molecule occupies an area of 3.01nm2. On the surface of the hanging mercury drop electrode, the adsorption of RUFX obeys Frumkin adsorption isotherm. The adsorption coefficient beta is 7.74X105, the number of electrons transferred (n) is 2, the transfer coefficient alpha is 0.76, the rate constant of surface electrode reaction ks is 0.23s-1. The mechanism of electrochemical reduction of RUFX at Hg electrode is explored. The detection limit of the adsorptive stripping voltammetry of RUFX is 1.0X10-9 mol/L under optimized condition. CONCLUSION: This indicates an irreversible process with adsorption.
