Practical methods, developed for transferring yeast cells under sterile, anaerobic conditions from their liquid growth medium to sugar-free media, provided "washed" cells which had lost only negligible amts. of intracellular carbohydrates, lipids, and riboflavin. Riboflavin was rapidly lost from washed cells suspended in sugar-free phosphate solns. The loss was much greater in the presence of oxygen than in its absence. Of the various concurrent processes involved in this loss, the best positive correlation was obtained with the process of vacuole formation. During aeration in M/15 KH2PO4 the increase in number of cells having enlarged central vacuoles paralleled in a general way the development of vit. B2 deficiency in each suspension; the "vacuolated" cells in these "dissimilated suspensions" had apparently lost most of their riboflavin. Cytological and chemical evidence indicated that suspensions of yeast cells in sugar-free phosphate solns. are made up of a mixed population of cells differing in riboflavin content and exhibiting different degrees of metabolic activity.
