Griseofulvin-warfarin antagonism


, : Griseofulvin-warfarin antagonism. JAMA 199(8): 582-583

The effect of griseofulvin on the anticoagulant action of sodium warfarin was seen in 4 subjects. Relatively constant anticoagulant levels with warfarin (Coumadin Sodium) were achieved before oral administration of griseofulvin. Anticoagulant activity was measured by daily prothrombin times. In 3 of the 4 cases, a significant depression of anticoagulant activity was returned to pretreatment levels on discontinuing the griseofulvin therapy. These findings demonstrate the need for careful monitoring of patients on anticoagulant therapy when given griseofulvin and also suggest the importance of close observation of these patients when other drugs such as ethchlorvynol are prescribed.
