Rat kangaroo cells treated with a high dose of .alpha.-amanitine showed swelling of nucleoli within a short period. The nucleoli disintegrated at the light microscope level when the cells are treated for a longer period. Light microscopic autoradiography indicated an inhibition of extra nucleolar RNA synthesis within a short period but grains were formed all over the nucleoplasm when cells are treated longer. This was due to nucleolar segregation and dispersion of different nucleolar components. EM studies revealed that the nucleolar fibrillar centers remained associated with the fibrillar components of the nucleoli. At the early stage of segregation the fibrillar centers, which remained embedded in the nucleolar mass in control cells, took up peripheral position but got scattered in the nucleoplasm with longer treatment.
