The teratogenic and embryo-toxic properties of sarcolysine ([image]-di-(2 -chloroethyl) -amino -dl-phenylalnine) with respect to rat embryos were studied. Sarcolysine was injected as physiological solution into pregnant females at specified times during pregnancy. The results were determined on the 17th day after fertilization. Implantation percentages were determined and embryos were examined in situ. In the 1st series of experiments, sarcolysine was injected daily at a dose of 10 mg/kg from day 1 to day 15 of pregnancy. A total of 44 percent of implanted embryos died and spontaneous abortion occurred on days 4 and 5. More than 2/3 of the embryos died after injections given on days 2, 3, 6, 7 and 8. Injection at later times only were less lethal. Surviving embryos had a variety of non-specific malformations, particularly after injection on days 6 and 9. Brain defects were associated with injections on the 6th day. The day of peak sensitivity in terms of embryo lethality was established to be the 4th day of pregnancy. Minimal lethal doses for each day were established. A 15 mg/kg dose during days 12-14 resulted in a mortality rate of 30 percent, but no visible anomalies.
