The inhibitory effects of hadacidin [an antibiotic from Peniclllium frequentans] on human tumors growing in embryonated eggs and on formation of adenylic and deoxyadenylic acids led to study of the drug in patients with cancer. Earlier studied showed that hadacidin given once daily caused unpredictable toxic effects but that fractional drug doses given orally at 6-hr, intervals, with attention to the renal function and analysis of serum, drug levels, led to more accurate prediction of the tolerable dose and duration of treatment. Tumor regressions were slight and infrequent among 98 adult cancer patients whose treatment was randomized between 3 different doses given every 6 hr. Further trials of hadacidin in patients with solid tumors are not indicated.
