The serum of rabbits injected intravenously with erythrocytes of several inbred strains of mice agglutinated and hemolyzed these erythrocytes in the test tube and produced anemia when injected into mice. These in vitro and in vivo activities increased progressively until the maximum titers were reached after from 3 to 4 courses of four biweekly injections. Thereafter, the in vitro titers were maintained or increased moderately, whereas the ability to produce anemia decreased steadily. This dissociation pattern was less distinct in splenectomized rabbits and still less distinct or even absent in sera of intraperitoneally injected rabbits. These observations suggest that the in vitro reacting antibodies are not necessarily a measure of the in vivo reactivity, especially on prolonged immunization. The relative in vivo potency of hemolytic antiserum compared with its in vitro reactivity was expressed as "hemolytic ratio". It is possible that the differences in the in vitro and in vivo reactivities are related to changes in gamma globulin fractions which we reported previously.
