A glucose solution was infused into 7 dogs over a 4-8 hour period. Variations in glycemia, arterio-venous sugar difference, glycosuria, blood-lactic acid, extracellular water volume sodium thiocyanate, and heart output were recorded directly, and glomerular filtration calculated indirectly. Glucose metabolism was measured by the arteriovenous sugar difference plus the hourly difference between amounts perfused on the one hand and either eliminated or present in the extracellular liquids on the other. Two different periods were noted. The first is characterized by concomitant increase in glycemia and assimilation yield, corresponding to a stimulation of the pancreatic cells. During the second phase two different behaviors were noted in some an increase in glycemia with lower cardiac output and glomerular filtration; in others oscillation of blood sugar about a plateau with cardiac output and glomerular filtration stable. However, a net reduction in assimilation was noted on the whole, due to lower rate of cellular penetration by the glucose, saturation reactions in the pancreas, and changes in cell metabolism.
