To determine whether the intratracheal method can be used for quantitative results, rats were injected with 50 mg of silica suspended in 1 ml saline, and the silica content, collagen content and corrected lung weights (weight of lung per 100 g body weight) were determined at intervals. It was found that only part of the inoculum is retained in the lungs but with a standarized technique this is sufficiently constant. Statistical analysis of the results showed that right and left lungs should be kept separate for the various investigations as a wide range of values is needed to establish correlation between the given data. It was confirmed that collagen content and corrected lung weight can be used as a measure of reaction of the lungs to silica, and the number of animals that would be required to ensure that the sample mean will fall within 10% of the unknown true mean in 95 out of 100 cases was determined.
