Im-plants of various materials were made under the frontal cortex, in the anterior end of the callosal radiation, of the rat brain. Certain inorganic and organic chemicals produced local necrosis and widespread edema of white matter associated with increased permeability of the blood-brain-barrier. Purified protein derivative of tubercle bacilli (PPD) and endotoxin also caused edema with increased permeability but in addition there was a dense neutrophilic exudate limited to white matter. Pneumococcal and cryptococcal polysaccharides resulted in severe vacuolation of white matter, probably restricted to the interfascicular oligodendroglia, but in this case there was little or no alteration of permeability. The occurrence of three different histo-logic patterns of injury with the same localization in white matter is explained on the basis of a specific mechanism in white matter that facilitates spread of various substances. These results may help to explain the selective localization of cerebral edema and demyelinating diseases in white matter.
