The experimental technique is described. McCollum's diet 3143 was used and x-ray examination was the chief method of determining the extent of calcification, though histological preparations were also made. At least 18 rats are required for a test. Of these 6 receive the rachitic diet continuously, 6 receive the rachitic diet and the material under observation, and 6 receive the rachitic diet and a preparation that is known to be an active antirachitic agent. A satisfactory procedure is to give all animals the Mc-Collum diet for 14 days. The material to be examined is then offered in addition for an 8-day period. Substances studied for antirachitic activity are dissolved in a suitable solvent, and given per os through a pipette. The daily dose of the substance under examination varied from 0.01 to 0.001 mgm. The following substances possessed antirachitic activity: irradiated ergosterol, the distillate of irradiated ergosterol, irradiated ergosterylacetate, irradiated digitaligenin and irradiated acetyldigitaligenin. The following substances were inactive, whether irradiated or not; isoergosterol, oxycholesterylene, C27H42O, nerolidol, and pseudoionone.
