Ethanol and acetate at concentrations of 2.5,10 and 60 mmolar decrease the radioactivity of the liver slice fatty acids significantly below the level found in control experiments in which only 0.1 m[image] sodium acetate- 1-C14 was added. The suppressive effect of ethanol becomes maximal at 10 m[image] whereas that of acetate increases with the concentration and becomes maximal at 60 m[image], at which point the suppression becomes almost complete. These results show that the suppressing effect of ethanol on the incorporation by labeled acetate into hepatic fatty acid can be explained as being mostly due to isotopic dilution of labeled acetyl-CoA by unlabeled acetyl-CoA derived from ethanol. These results also show that the claim (Lieber and Davidson, Am. J. Med., 33, 319, 1962) that ethanol stimulates incorporation of acetate into fatty acids is based upon the interpretation of inadequately controlled experiments.
