Fatty acid synthesis in mitochondria. Elongation of short-chain fatty acids and formation of unsaturated long-chain fatty acids

, : Fatty acid synthesis in mitochondria. Elongation of short-chain fatty acids and formation of unsaturated long-chain fatty acids. Biochimica et Biophysica Acta 152(1): 50-62

The rate of incorporation of [14C]acetyl-CoA, [14C]malonyl-CoA and [14C]-pyruvate into fatty acids by rat heart sarcosomes has been studied. Under aerobic conditions acetyl-CoA and malonyl-CoA are incorporated at about equal rates, whereas pyruvate is incorporated most rapidly. Under anaerobic conditions acetyl-CoA incorporation is stimulated considerably, while malonyl-CoA incorporation is stimulated only slightly, which suggests that the substrate actually incorporated is acetyl-CoA and not malonyl-CoA. The incorporation of acetyl units appears to be strictly dependent on the presence of either endogenous or added fatty acids.
