The in vitro susceptibility of mouse peritoneal macrophages to ectromelia virus was determined by using immune fluorescence to detect infected cells. Macrophages were more readily infected by virulent virus than by attenuated virus. Strains of mouse which differed in their susceptibility to mousepox nevertheless gave macrophages which were equally susceptible to infection. Macrophages from mice which were immune to ectromelia were more susceptible than were macrophages from normal mice even with immune serum. This increased susceptibility is attributed to the greater surface area and phagocytic activity of macrophages and is not specifically directed against ectromelia virus.
