Glyoxal down to 0. 5 mM is cytotoxic for human fibroblasts in culture by blocking DNA and protein synthesis primarily. The synthesis of rapidly-labelled RNA is not inhibited by short treatment even with high doses of glyoxal; that of ribosomal RNA is suppressed almost entirely after a 5-h treatment, but no change was observed in the sedimentation profile of the main components. The activity of thymidine kinase fluctuated during the cell cycle, though glyoxal had no effect on the enzyme in vitro. It is assumed that glyoxal inhibits DNA replication; transcription is inhibited only in the later phase of action.
