Aqueous solutions of apomorphine hydrochloride are unstable, turning green within 10-15 min. and depositing dark-colored sediment after 4-5 days. These changes were not prevented by protection from light. Nor were they prevented by adding either ethylenediamine tetra-acetate (EDTA) or ascorbic acid to the water prior to addition of apomorphine. Methanol, ethanol, and 10% ethanol solutions were no more permanent than water. Low pH and absence of O2 were found to favor stability. Apomorphine solutions buffered at pH 1-3 did not change color over a 6-month period. They changed color at pH 4.8 and higher, and the amount of change increased with increase in pH and time. At 37[degree]C and pH 5.8, 98.0% was unchanged after 2 hr. and 85.4% unchanged after 8 hr.; at pH 7.9, 71.2% was unchanged after 2 hr. and 18.4% after 8 hr. Changes did not occur in pH 5.5 solutions during 40 hr. when the water used in preparing the solutions had had O2 removed by evacuation and nitrogen flushing (Thunberg tube). The green-colored decomposition product lacks the emetic and central nervous stimulating effects of apomorphine in dogs; it was mildly sedative to these animals.
