Rabbit lung slices oxidized glucose to CO2 and converted it to glycerol and fatty acid moieties of glycerides. The ratio of glucose-1-C14 to glucose-6-C14O2 was 2.5. The oxidation of glucose was not affected by insulin. Palmitate did not affect oxidation of glucose but increased its incorporation into the glycerol moiety of the glycerides while inhibiting lipogenesis from glucose. Glucose stimulted lipogenesis from acetate. Most of the glucose and palmitate was incorporated into the phospholipid fraction, primarily lecithin, of the lipids. Increase in palmitate concentration in the incubation medium, up to 1.0 [mu]mole/ ml, increased the incorporation of palmitate into lecithin. Exogenous choline was incorporated into lecithin but did not significantly affect the rate of palmitate incorporation. The active synthesis of lecithin by lung slices probably is related to the formation and secretion of surfactant by the lung.
