Congo red, benzoazurine, azo blue, and several other dyes of this type can be separated by means of a H2O-amyl alcohol mixture into hydrophilic and lipophilic components. Each component shows a characteristic absorption spectrum. Cellulose membranes are colored exclusively by the hydrophilic component, lignified or cutinized membranes by the lipophilic component. The view that such metachromatic staining is due to a dye of one definite compound which produces different shades of color in different parts of the cell is untenable. Measurement of the size of submicroscopic membrane pores by use of such dyes is not possible. The hydrophilic or lipophilic nature of certain cell-wall components may be thus detd.
