Tritium-labeled water and tritium-labeled cholesterol were used to investigate several aspects of endogenous and exogenous cholesterol metabolism in exptl. atherosclerosis. The metabolic turnover of cholesterol in the normal rabbit aorta was demonstrated by the presence of labeled cholesterol in the aorta following a period during which the body water had been labeled. Most of the cholesterol deposits in the atherosclerotic aorta of 2 rabbits fed tracer cholesterol for 1 mo. were of exogenous origin. The presence of endogenous cholesterol in small amts. also was found in the atherosclerotic aorta. Tritium-cholesterol was fed in single doses to 4 rabbits and the rise in specific activity in the serum cholesterol observed. A max. specific activity was found in from 2 to 4 days. The turnover rate of esterif ied cholesterol of the serum was found to be faster than free cholesterol. Labeled cholesterol was found in all tissues examined following an oral dose of about 0.15 mc. of tritium-cholesterol.
