Quantitative glycogen determinations in blood

Differing results were obtained in comparing the data on blood glycogen content determinations by means of reducto-metric (Hagedorn - Jensen) anthrone and enzymatic (glucoseoxidase) methods. Galactose, mannose and an unidentified monosaccharide were demonstrated chromatographically in hydrolyzates of preparations investigated. Reductometric and antrone methods therefore could not oe used for glycogen content determination in whole blood, whereas for glycogen content determinations in leucocytes each of 3 methods, mentioned above may be applied. Glucose content values in glycogen hydrolizates determined by glucoseoxidase method and by means of paper chromatography were found to be identical. A method for quantitative glycogen determinations in whole blood by means of commercial glucoseoxidase preparation is described.
