Research on capillary permeability activity of 52 synthetic peptides related to eledoisin and physalaemin

, : Research on capillary permeability activity of 52 synthetic peptides related to eledoisin and physalaemin. Arch Int Pharmacodyn Therap 160(1): 44-54

Fifty two synthetic peptides related to eledoisin and physalaemin were screened for their ability to increase the permeability of the skin vessels in man and guinea-pig. There is evidence that: reduction of the size of the eledoisin and physalaemin molecule by means of a progressive elimination of the N-terminal amino acid residue reduces edema-provoking activity of the peptides; the N-terminal lysine residue yields inactive peptides, whilst the N-terminal asparagine residue leaves unaltered or actually increases capillary permeability. A marked difference between hypo-tensive and oedema-provoking activity was observed in these 2 groups of compounds; any change in the three amino acids of the C-terminal moiety of the eledoisin and physalaemin molecule (-gly-leu-met-NH2) and substitution of phenylalanine residue yields inactive peptides.
