Plasma thromboplastin antecedent (PTA) and Hageman factor (HF) have been separated from human plasma by ion exchange chromatography using diethylaminoethyl cellulose resin. A constant pH of 7.12 was used; salt concentration was increased continuously from 0.05 [image] to 0.3 [image] by varying NaCl concentration in presence of 0.02 [image] sodium diethyl barbiturate buffer. The maximum PTA activity concided with the first protein peak while the greatest HF activity immediately preceded the second protein maximum. Each fraction contained less than 1% of normal of all other known clotting factor activities. Each purified fraction corrected the defect of its corresponding deficiency plasma. However, the combined purified factors failed to shorten the clotting time of "exhausted plasma" (prepared by Waaler's technique of treating normal plasma with celite powder) unless another as yet unidentified component of plasma was also present.
