Methyl bis (2-chlorethyl) amine was administered intravenously to Dutch and New Zealand strain rabbits and Sprague-Dawley and Long-Evans rats to evaluate the effects of N mustard upon the antibody and/or cellular factors involved in homograft rejection. In rats, 2 dosage schedules were used following reciprocal cross-grafting between 5 Sprague-Dawley and 5 Long-Evans rats and between 20 Sprague-Dawley rats. Homograft survival, determined by daily gross observation and serial biopsy of grafts, was compared between treated and comparable untreated groups. Significant leukopenia was not achieved in rats but marked weight loss and lymphoid tissue atrophy was noted. Three dosage schedules were used in groups of 10 rabbits composed of equal numbers of both strains. In all treated rabbits, marked leukopenia was confirmed by daily individual leukocyte counts. Marked weight loss and lymphoid atrophy was noted also. There was no significant prolongation of homograft survival in the treated rat and rabbit groups over that noted in comparable groups of untreated animals. These data indicate that intensive N mustard administration does not depress host immune mechanisms to the degree that homograft survival is prolonged. This is contrary to the published observations of others. Also, it appears that large numbers of circulating host leukocytes are not necessary for homograft rejection to occur.
