Ultrastructure of Botrydium granulatum

The fine structure of the multinucleate alga B. granulatum was examined by electron microscopy. In these cells the different energids are not separated by membraneous structures. The plastids are (in addition to their own double membrane) surrounded by an outer envelope which is sometimes continuous with cisternae of the endoplasmic reticulum or, especially during the formation of zoospores, connected with perinuclear cisternae. Bands, consisting of 3 thylakoids each, traverse the plastids. Within these bands the thylakoids are not tightly appressed, but loosely associated with a distinct, narrow space between them. The plastids contain pyrenoids as regions of a somewhat denser matrix which are not limited by particular membranes. In each energid there is a Golgi apparatus composed of 2 reticulate dictyosomes, which are located in a typical parabasal position, relating to the 2 centrioles or the flagellar bases, respectively. An association between these dictyosomes and the nuclear envelope is apparent. Vesicles, which seem to derive from blebs of the outer membrane of the nuclear envelope, appear to coalesce with the cisternae of the dictyosomal forming face. Furthermore, evidence is presented for the derivation of some characteristic vesicles like the "coated" and "fusiform" vesicles from the dictyosomes. The structural features are discussed with reference to the position of the Xanthophyceae in the taxonomic system of algae.
