A contribution to fine structure of chloroplast lamellae

, : A contribution to fine structure of chloroplast lamellae. Protoplasma 64(2): 185-194

Lamellae within the grana and stroma of tobacco and soybean chloroplasts were studied in the electron microscope after fixation with 0.6% KMnO3. Stroma lamellae consisted of a single row of spheroidal subunits. Grana lamellae contained 2 chains of spheroidal subunits, with a radius of about 45 A each. The subunits contained protein and lipids. The absence of a continuous dark rim as well as the electrophoretical behavior of lamellae fragments suggested the presence of protein on the surface of both the subunits and the lamellae themselves. Distribution of lipids in the subunits was discussed.

In 6-hydroxy-D-nicotine oxidase (6-HDNO) FAD is covalently bound to His71 of the polypeptide chain by an 8 alpha-(N3-histidyl)-riboflavin linkage. The FAD-binding histidine was exchanged by site-directed mutagenesis to either a Cys- or Tyr-residue...
