The partition of phosphate in trichloroacetic acid (TCA) extracts of mold tissue can be correctly assessed only by adopting the method of Berenblum and Chain, Biochem. J. (London) 32, 295 (1938), as modified by later workers, where the phosphomolybdate is extracted into isobutyl alcohol, prior to color development. Conventional methods, which attempt to develop the color directly on the extracts, can lead to considerable error owing to the presence of interfering material which reacts with molybdate and with reducing agent.
