In human liver homogenates the conjugation of C14-labelled cholic acid was studied by thin-layer chromatography (TLC). Well-defined specific areas of taurocholate, glycocholate and free cholic acid were identified after spraying with water under transillumination. These areas were scraped into thixotropic gel and their radioactivity was measured in a liquid scintillation spectrometer. The results were compared with those from standard paper chromatography and direct determination of radioactivity in an automatic G.M. counter with mica-end window. Both methods gave the same final results. (The 2 methods were run parallel and all experiments were duplicated.) The TLC method permits considerable saving of time (2-3 days) in serial determinations. The TLC method also gives appreciably greater reliability in calculation of results, since recovery of the added radioactivity was more complete.
