The growth inhibition by 8-azaguanine of T. geleii H. a ciliate which requires guanine, which it converts to adenine irreversibly, was found to be as readily released by adenine and hypoxanthine as by guanine, at low inhibitor concns. However, when T. geleii was grown with optimal amts. of both adenine and guanine-8-C14, the extent to which nucleic acid adenine was derived from each was the same in the absence as in the presence of low inhibitory levels of 8-azaguanine, thus failing to substantiate the hypothesis of a specific block by the azaguanine. Of 3 potential intermediates tested, xanthosine and 4-formamidino-5-imidazolecarboxamide were nutritionally inert. Hypoxanthine-8-C14 and adenine contributed equally to the nucleic acid adenine of T. geleii, when the ciliates were grown with these and suboptimal guanine, but the hypoxanthine was not converted to nucleic acid guanine. The ratio of bound adenine to guanine in the organisms was found to be 1.1:1. The total nucleic acids were calculated from the bound purine content to be 4.5% of the dry wt., from colorimetric sugar tests to be 4.9%. The ratio of desoxyribose nucleic acid to ribose nucleic acid was 0.10.
