The antimicrobial activity of saponin fraction from the roots of <i>Hemidesmus indicus</i> was evaluated against pathogenic bacteria and fungi in an <i>in vitro</i> condition by agar diffusion assay. Pure saponin extract exhibited remarkable antimicrobial activity against <i>Staphylococcus aureus</i>, <i>Salmonella typhi</i>, <i>Klebsiella pneumoniae</i>, <i>Aspergillus flavus</i>, <i>Aspergillus fumigatus</i> and <i>Aspergillus niger</i>.
