kappa-Casein (kappa-CN) is the milk protein that determines the size and specific function of milk micelles, and its cleavage by chimosin is responsible for milk coagulation. Recently, a method was published describing a simultaneous genotyping procedure that detects seven goat kappa-CN variants, using the primer extension method. Until then, goat kappa-CN variants genotyping was mainly done by time-consuming methods (like the PCR-Restriction Fragment Length Polymorphism, RFLP procedure).Following this method, we started to genotype our local Portuguese goat breeds (Serrana, Bravia, Serpentina, Algarvia and Charnequeira), and preliminary results showed Alleles A and B as being the most frequent variants with the highest prevalence of the B variant (data in agreement with previous reports in other European breeds). The specific changes we did to the original protocol, at the PCR protocol, are shown in this report, and where done using a Gradient PCR device.
