Objective: To evaluate salt intake during the perinatal period on IS and BP in adult Wistar rats.Methods: Female Wistar rats were fed a low (LSD: 0.15% NaCl), normal (NSD: 1.3%), high1(HSD1: 4%) or high2 (HSD2: 8%) salt diet since 8 weeks of age. At 12 weeks of age, they were matched with males that received only NSD. Weekly tail-cuff BP (tcBP) was performed during pregnancy. Only 8 pups (4 males/4 females) were kept with their mothers and, in the remaining, neonatal glucose levels (NGLUC) were measured. After weaning, all offspring received only NSD. At 12 weeks of age, intra-arterial mean BP (MBP) was evaluated, and a euglycemic hyperinsulinemic clamp (CLAMP) was done.Results (mean(SD): During pregnancy tcBP was higher (p<0.001, n=4/group) on HSD2 (142+-9.6) and HSD1 (130+-8) than on NSD (124+-11) and LSD (112+-8 mmHg). NGLUC was lower (p<0.05) in male offspring from LSD dams than in the other groups (HSD2 76(10, n=4; HSD1 84(4, n=4; NSD 62(7, n=13; LSD 51(2.5 mg/dl, n=4). IS was lower (p<0.01) in male offspring (n=6/group) from LSD dams (HSD2 16.5(4, HSD1 14.7(3, NSD 10.8(3, LSD 3.6(0.7 mg.kg.min-1). Higher MBP (p<0.05) was observed in male and female offspring (n=6/group) from HSD2 and HSD1 dams (males: HSD2 110(5, HSD1 107(4, NSD 100(2, LSD 92(2; females: HSD2 112(4, HSD1 110(3, NSD 103(3, LSD 98(4).Conclusions: NGLUC, and IS and BP in adults are influenced by maternal salt intake during pregnancy and lactation.
