Cadmium (Cd), an ubiquitous heavy metal, has been known to damage the immune system including T lymphocytes, although their mechanism was not fully elucidated. In the present study, we tried to investigate the signaling pathways involved in the Cd-induced cell death in human T cell line, Jurkat cells. Cell viability was determined by MTT assay. The treatment of 3-30μM Cd induced cell death in a dose-dependent manner. Indeed, in the cells treated with 10 μM Cd, cell viability was about 60 % compared to control. To evaluate whether this cell death includes apoptosis, we performed Annexin V staining by FACScan. Annexin V stained cells were dose-dependently increased in Cd-exposed Jurkat cells. To investigate the signaling pathway for apoptosis, the effects of various inhibitors for MAPKs and PI3 kinase on Cd-induced apoptosis were examined. In MTT assay, wortmannin, an inhibitor for PI3K, abolished the Cd-induced cell death. In addition, Cd-induced cell death was completely blocked by SB202190 (an inhibitor for p38 MAPK) and PD98059 (an inhibitor for MEK), but not by SP600125 (a specific inhibitor for JNK). In summary, we suggest that Cd-induced apoptosis may involve PI3K/p38 MAPK signaling pathway. This study is supported by Ministry of Environment as ′The Eco-technopia 21 project′.
