We have shown that dietary long chain n-3 polyunsaturated fatty acids (PUFA) present in fish oil (FO) affect CD4+ T cell proliferation and cytokine production in C57BL/6 mice. We hypothesized that FO might alter the balance between Th1 and Th2 subsets. To elucidate the effect of FO on Th2 polarization, mice were fed a wash out control diet (5% corn oil (CO), n-6 PUFA) for one week, followed by the same control diet or a fish oil diet (1% CO + 4% FO, n-3 PUFA) for 2 weeks. Splenic CD4+ T cells were polarized toward a Th2 cytokine profile in vitro by culturing with anti-CD3 and PMA, rIL-4, anti-IFN-? , and rIL-2 for 2 days. Cells were reactivated with PMA and ionomycin in the presence of monensin for assessment of expression of the prototypic Th2 and Th1 cytokines, IL-4 and IFN-? , respectively, using intracellular cytokine staining and flow cytometric analysis. Approximately 20% of the CD4+ T cells were polarized to Th2 cells, as indicated by positive staining with anti-IL-4-PE and negative staining with anti-IFN-? -FITC. In addition, the proliferation of Th2 cells was measured by 3H-thymidine uptake. The FO diet did not significantly affect proliferation or cytokine production of the polarized Th2 cells compared with the CO diet. Thus, the beneficial effects of FO do not appear to be explained by a shift in the propensity of CD4+ T cells to be polarized toward a Th2 phenotype, at least in vitro.
