Newly discovered galanin-like peptide (GALP) is a 60 amino acid neuropeptide. Amino acids 9 to 21 of GALP are homologous to the first 13 amino acids of galanin. GALP is a member of GPCR ligand and reportedly implicated in neuroendocrine regulation of body weight and food intake. We found that GALP is expressed in neurons in the arcuate nucleus (ARC) and nerve terminals in the external layer of the median eminence. Injection of GALP itself into the paraventricular nucleus appears to stimulate the food intake. Neuronal mechanisms of feeding regulation remained unclear. The aim of this study is to determine which area and what kind of neurons are activated by GALP. In this study, we used male Wistar rats and either GALP (3 nmol) or vehicle (saline) was injected into the lateral ventricles. The animals were fixed by fixative containing 4% paraformaldehyde 2-h after the injection. Fresh frozen sections were prepared and then stained with a double fluorescence immunohistochemistry using antibodies against c-Fos, tyrosine hydroxylase (TH), orexin and melanin-concentrating hormone (MCH). As results, we confirmed that intracerebroventricular (icv) injection of GALP increased food intake. GALP induced Fos-positive cells in the periventricular zone of the third ventricle. Some TH-positive neurons expressed Fos-immunoreactivity in the ARC. However, orexin-or MCH-positive neurons did not show any Fos-immunoreactivity after icv injection of GALP. It is possible that feeding regulation by GALP does not mediate through orexin and MCH. In conclusion, GALP is a novel orexigenic peptide and may stimulate food intake through TH-containing neurons in the hypothalamus.
