Our previous work shows that stimulation of the sciatic nerve results in GLU release in the hindpaw. This study investigates release of GLU and ASP in the dorsal root ganglia (DRG) following hindpaw stimulation. One hindpaw of normal male SD rats (280-340g) was injected with 100mul Complete Freund's Adjuvant (CFA) or phosphate buffer saline (PBS). Five days later, a microdialysis fiber was placed in the ipsilateral lumbar 5 DRG. Following 120min washout period (2mul/min), GLU and ASP baselines were established by sampling every 10min, then the hindpaw was exposed to thermal (23degreeC or 52degreeC water) or mechanical (brushing) stimuli for 20min while collecting dialysis samples. Compared to normal DRGs (n=4), baseline GLU and ASP in inflamed animals (n=4) was increased 74% and 19%, respectively. Compared to baseline, a 23degreeC stimulus did not increase GLU or ASP in the DRG of CFA animals (n=3), but a 52degreeC stimulus resulted in a peak increase of 70% and 95% in GLU and ASP, respectively in PBS animals (n=4) and a 212% and 188% peak increase in CFA animals (n=4). Following mechanical stimulation, GLU and ASP did not change in the PBS group but a peak increase of 27% and 144% in GLU and ASP respectively, occurred in DRGs of CFA animals (n=3). The results indicate that GLU and ASP are released in DRGs following noxious thermal stimulation in normal rats with a dramatic increase in inflamed animals. Innocuous mechanical stimulation also exacerbates EAA release in the DRG in inflamed rats. Thus, EAA release in DRGs may contribute to allodynia and hyperalgesia in inflammation.
