Oxidative stress has been proposed as a mechanism for age-related alterations in brain function and in neurodegenerative diseases such as Alzheimer's disease and Parkinson's disease. This state of oxidative stress induces perturbations in DNA, proteins, and fatty acids. We have shown that protein levels of the bcl-2 family of proteins is upregulated in the aged brain and that antioxidnat treatment returns protein levels to normal. This alteration in bcl-2 family proteins in the aged brain likely represents a response to prolonged oxidative insult. Prolonged exposure to normobaric hyperoxia (>99%O2, 1 ATM) is an oxidative insult that can mimic some age-related neural deficits, including alterations in bcl-2 family proteins. In this investigation we examined gene regulation of apoptotic proteins using the ribonuclease protection assay. Expression of Bax, Bcl-X, Bcl-2, caspase 1, 2, and 3, FAS and FAS ligand were measured in young, aged, and hyperoxia exposed young F344 rat brain. Increases in Bcl-X and Bcl-2 mRNA were observed in the cerebellum of the aged rats, suggesting that alterations in bcl-2 family proteins may be reflected in a change in gene regulation.
