We have previously shown that white matter glia in situ respond to ATP with an increase in (Ca2+)i, and that this response is mediated by both P2Y and P2X receptors. We suggest that the low-affinity component of the ATP response could be mediated by the pore-forming subtype P2X7, which has important roles in pathophysiology. In this study we investigate whether white matter glia in situ possess the P2X7 subtype in intact isolated optic nerves of postnatal day 10-30 rats. Rats were killed by CO2 narcosis and optic nerves were removed. Intact optic nerves were then either calcium imaged or snap frozen and sectioned for immunohistochemistry. Application of ATP caused a concentration-dependent increase in glial (Ca2+)i, with an initial response at 10 nM ATP and a maximal response at 10 mM ATP ((Ca2+)i increase = 123 +- 10 mu M), with an EC50 of 25 mu M ATP. The response to 1 mM ATP was potentiated by removal of Na+ and Mg2+ from the extracellular milieu by, respectively, 160 +- 20% and 46 +- 11%, consistent with the effects of these ions on P2X7 receptors. The P2X7 receptor agonist BzATP caused a concentration dependent increase in (Ca2+)i, with a maximal response at 1 mM BzATP ((Ca2+)i increase = 43 +- 2 mu M; EC50 = 32 mu M ATP), and this could be blocked (100 %) at 30 mu M by the P2X7-specific antagonist, periodate-oxidised ATP (100 mu M). Immunohistochemistry confirmed expression of P2X7 receptor in optic nerve astrocytes, oligodendrocytes and microglia. In summary, the results demonstrate expression of P2X7 receptors in optic nerve glia in situ, and we suggest that these and other purinoceptors may have important roles in the pathophysiology of white matter.
