p62, a cellular protein which was initially identified as a phosphotyrosine-independent ligand of the SH2 domain of p561ck, has been known to interact with several proteins such as atypical protein kinase C(aPKC), RIP and TRAF6. Here, we demonstrate that p62 interacts with prostate apoptosis response-4 (PAR-4). PAR-4 has been shown to interact with PKC-zeta and negatively regulate its kinase activity. The p62-PAR-4 interaction was verified by co-immunoprecipitation in HeLa cell lysate and by GST-PAR-4 pull-down assay in vitro. As p62 is known to interact with PKC-zeta, p62 might form a ternary complex with PAR-4 and PKC zeta. This ternary complex was confirmed by simultaneous co-immunoprecipitation of these three components both in the HeLa cell lysate and in the vitro transcription/translation products. The patterns of these interactions and the biological meaning of the ternary complex will be discussed.
