Role of AT1 angiotensin receptor in the tonic and reflex control of blood pressure in renovascular hypertensive rats

It is well known that angiotensinergic system is involved in the generation and maintenance of high blood pressure (BP) in renovascular hypertensive model. The aim of the present study was to elucidate the importance of angiotensin AT1 receptor in the tonic and reflex control of BP. Wistar male rats were distributed in 4 groups: 1) a six weeks Goldblatt 2 kidney-one clip (2K1C) group (n=6), 2) a 2K1C group treated with Candesartan Cilexetil (10 mg/Kg/day) for three weeks (n=6), 3) a 2K1C group treated with captopril (50 mg/Kg/day) for three weeks (n=6), and 4) a control no treated group (n=6). Twenty four hours before the experiment, animals were anesthetized with ketamine-xylazine (40mg/Kg and 20mg/Kg respectively) for femoral artery and vein cannulation in order to measure BP and heart rate (HR) and drugs (L-Phenyllefrine and Sodium nitroprusside) administration respectively, and to evaluate the baroreflex. The BP level in group 1 was (18910.9 mmHg); group 2 (1112.6 mmHg), group 3 (12810 mmHg) and group 4 (992.7 mmHg). The sympathetic component of baroreceptor evaluated by nitroprusside was: in group 1 (-1.80.3 bpm/mmHg); group 2 (-3.740.2 bpm/mmHg), group 3 (-3.520.6 bpm/mmHg) and group 4 (-3.240.2 bpm/mmHg). And finally, the parassympatetic component was in the group 1 (-1.920,2 bpm/mmHg); group 2 (-2,370.2 bpm/mmHg), group 3 (-2.270.2 bpm/mmHg) and group 4 (-3.280.2 bpm/mmHg). In conclusion, the results shows: 1) the high BP in the six weeks renovascular hypertensive model is dependant of AT1 receptors integrity, 2) the impaired baroreflex control observed in this model is mediated by angiotensin acting on AT1 receptors.
