Proper synapse formation is a crucial component in maintaining efficient neurotransmission in the central and peripheral nervous system. During neurotransmission, receptors on the post-synaptic partner precisely cluster beneath the site of transmitter release. Rapsyn, receptor associated protein at the synapse, is a post-synaptic peripheral membrane protein that is a required component in clustering acetylcholine receptors at the neuromuscular junction. Rapsyn also clusters GABAA receptors and other synaptic molecules in heterologous cells. Our objective is to determine which domains of rapsyn are responsible for clustering synaptic proteins. To this end, we have generated rapsyn deletion constructs and GST-rapsyn deletion constructs. These mutants have been shown by Western analysis to express mutant rapsyn proteins of the expected molecular weight. Using immunofluorescence, we have found that specific domains of rapsyn affect its ability to self-associate and interact with other proteins. These data suggest that particular domains of rapsyn aid in its ability to cluster synaptic molecules.
