Mice homozygous for the Stat4-/- mutation were sensitized to cockroach antigen and challenged intra-tracheally 21 days later. Surprisingly, the Stat4-/- mice showed significant decreases in airway hyperreactivity and peribronchial eosinophils compared to wildtype (wt) controls. In addition, pulmonary levels of Type 2-like chemokines were decreased in the Stat4-/- mice, including CCL1, CCL5, CCL6, CCL11, CCL17 and CCL22. However, levels of cytokines such as IL-4, IL-5 and IL-13 as well as serum IgE levels were similar in the two groups. Transfer of splenocytes from wt mice into Stat4-/- mice partially restored airway hyperreactivity in the mutant mice. However, chemokine production and peribronchial eosinophilia was not restored. We hypothesize that Stat4 expression in cells other than lymphocytes or monocytes is required in this model for airway hyperreactivity and peribronchial eosinophilia to occur. Although an IL-12-responsive gene, it appears that Stat4 expression contributes to a Type 2 process such as allergen-induced airway hyperreactivity.
