Neuroprotection in focal stroke models has been achieved with selective inhibitors of COX-2 and also in COX-2 null mice. To elucidate COX-2 expression in ischemic brain, we performed permanent middle cerebral artery occlusion in adult Swiss-Webster mice. Brains were prepared for Western blotting and immunohistochemistry 6 or 24 h later. COX-2 protein expression increased at 6 h postischemia in penumbra and core, and remained elevated at 24 h. At 6 h, perinuclear staining was evident in neurons and microvessels of the cortical penumbra; by 24 h, the number of immunoreactive neurons and microvessels in the penumbra was increased considerably, as was the intensity of the labeling. In the ischemic core, moderate staining of microvessels was evidenced, with occasional immunopositivity in surviving neurons. By 24 h, when widespread neuronal death had occurred, the frequency and intensity of microvessel staining was increased, and neuronal immunopositivity was no longer evident. These findings indicate that COX-2 is induced rapidly and progressively in neurons and vascular cells after focal stroke. The relative contribution of these cells to the COX-2 induced brain injury remains to be clarified.
