Stimulation of laryngeal mucosal afferents elicits protective adductor reflexes in the cat larynx. To determine the transynaptic pathway involving these afferents in the medulla, we used the Becker strain of pseudorabies virus (PRV) because of its differential tropism for sensory neurons in the cat. Immediately prior to a unilateral mucosal PRV injection, the opposing superior laryngeal nerve (SLN) and ipsilateral recurrent laryngeal nerve (RLN) were sectioned to isolate the ipsilateral SLN afferents for PRV transport to the brain stem for 6, 11 and 14 day survival times. In another animal, we sectioned the ipsilateral SLN and RLN and left the contralateral SLN intact 10 days post injection. After immunohistochemistry, blinded counts of PRV labeled neurons were made at 7 levels (Berman, 1968) (16 caudal to 12.1 mm rostral) in 17 regions outlined using adjacent nissl stained sections. ANOVAs compared counts ipsilateral and contralateral to the intact iSLN, at each level across survival times.
