In this set of experiments, rats underwent unilateral sciatic nerve cut or compression (100 mmHg) and a sub-group was fed 50mg/kg/day of a standardized Ginkgo biloba extract post-op for either 1, 4, 8 or 12 wks. After euthanization, single fast-twitch extensor digitorum longus (EDL) fibers were dissected and demembranated (0.5% Triton X-100) and sensitivity to inorganic orthophosphate (Pi) and to calcium was assessed. Experiments were run at 22degreeC, pH 7, ionic strength of 200 mM, solutions formulated by solving the set of simultaneous equations describing the multiple equilibria of ions in solution and contained (mM): 5 EGTA, 20 imidazole, 2 Mg2+, 5 MgATP, 15 phosphocreatine and 100 u/ml creatine kinase. Appropriate Ca2+ and monopotassium Pi (KH2PO4) was added. Results indicate that nerve cut causes an initial large increase in Pi sensitivity (less force generated) and a decrease in Ca2+-sensitivity, both of which gradually recover, though not to control levels. Compression yielded an increased Pi sensitivity, and a decrease in Ca2+-sensitivity, nearly in direct proportion to the duration of compression. In most cases administration of Ginkgo biloba led to an amelioration of Pi sensitization and increased Ca2+-sensitivity back towards normal levels, providing evidence that muscle function may be maintained by antioxidant therapy.
