The therapeutic properties of cis-dichlorodiammineplatinum(II) (DDP) were studied in 12 strains of transplantable tumors and leukemias of mice. This compound possesses a wide spectrum of antitumor action. Under its influence, the growth of solid tumors of certain strains is markedly inhibited. The best effects were obtained in treating adenocarcinoma of the large intestine (strain AKATOL), cancer of the forestomach (strain PRG), sarcoma 180 and mammary adenocarcinoma 755 (Ca 755). The lifespan of mice with leukemia (strains L1210 and La), plasmacytoma MOPC-406 and ascitic hepatoma 22 was considerably increased. Animals with L1210 leukemia were in individual instances, completely cured. In 3 tumor strains (AKATOL, Ca 755 and sarcoma 37) a greater antitumor effect was obtained when DDP was given in combination with sarcolysine than when the drugs were used individually. Enhanced antineoplastic action was especially noticeable in the increase in the number of completely regressed tumors of strain AKATOL and sarcoma 37. Histologic studies of Ca 755 during chemotherapy showed that immunocompetent cells of the host play a prominent role in the mode of antineoplastic action of the Pt compound. This is manifested by the development of an intense lymphohistiocytic reaction around and inside the treated tumors. The main side effects of the drug are lesions of the convoluted tubules of the kidneys and villi of the small intestine.
